For optimal outcomes, it is recommended to run the device for five minutes. The plasma will be separated into five layers under centrifugal force, which are PRP, PPP (platelet-poor plasma), leuco-platelet layer, red fraction and separating gel. PPP is a clear fluid layer while leuco-platelet layer is a thin buffy coat comprised of white blood cells and platelets. The red fraction is made of red blood cells. PRP should be extracted with a syringe. Ideally, a centrifuge with a fixed-angle rotor (e.g. Plasmolifting Centrifuge XC 2415) should be used for PRP preparation.
